Overexpression of P-40 Anx-I ; confers low level drug resistance to structurally dissimilar anti-cancer drugs. Given the relatively low levels of drug resistance, it was of interest to examine the effects of inhibition of P-40 expression on drug resistance. P-40-expressing SKOV-3 cell line was transfected with the full-length reverse cDNA of Anx-I cloned into pCIN4 eukaryotic expression vector. Stable transfectants of reverse P-40 SKOV-3 cells were selected in the presence of lethal concentrations of G418. Fig. 4A shows a Western blot analysis of total cell extracts from SKOV-3 clones 16 and 17 probed with IPM96 mAb. As evidenced, clone 16 showed reduced expression of P-40 while clone 17 was completely depleted of the protein. Although Anx-I has no significant similarity with its family members at the nucleotide level, all annexins have a core domain that contains four homologous conserved repeats [21]. It was therefore of interest to verify that other members of the annexin family were not depleted simultaneously with Anx-I. Fig. 4A shows that neither Anx-II nor Anx-IV protein levels were affected in clones 16 and 17. To determine if the depletion of Anx-I affects the sensitivity of SKOV-3 cells to anti-cancer drugs, increasing concentrations of adriamycin, melphalan, and etoposide were assessed on the growth of SKOV-3 cells transfected with vector alone or reverse P-40 Fig. 4B ; . Clones 16 and 17 showed a 2.5to 8-fold decrease in their EC50 to adriamycin, melphalan, and etoposide relative to cells transfected with vector alone. Tissue through CIS to the malignant stage of a germ cell cancer, we performed a systematic comparison by the DDRTPCR method Liang and Pardee, 1992; Jorgensen et al., 1999 ; . For the initial screening, we analysed six testicular biopsy samples: two with homogeneous classical seminoma, two containing almost exclusively CIS tubules, of which one was from an intra-abdominal CIS and two with normal spermatogenesis Table I ; . We included the specimen of undescended testis with CIS because according to the hypothesis of testicular dysgenesis syndrome Skakkebaek et al., 2001 ; , both testicular cancer and undescended testes are symptoms of maldeveloped gonads and may have common pathogenesis. We performed 402 primer combinations, and assuming that each competitive PCR reaction gives rise to ~100 bands on a DDRT gel and that each band corresponds to one mRNA, this investigation allowed comparison of ~40 000 mRNA. We excised and sequenced 56 bands, which showed overexpression ranging from 1.5- to 10-fold ; in at least one of the CIS samples. Several mRNA were detected with more than one primer combination; for instance, overexpression of CCND2 was detected with three different primer combinations.
